Since the familiar is in the backpack, there is no line of effect from the source of the effect to the familiar, so it is not affected. GMs call but a backpack doesn't seem to pass muster as a solid physical barrier per the written guidelines. I wouldn't do it to a player, though, if the familiar is "stored" in a pouch or backpack.
